For many years, the Faculty of Medicine was the only functioning faculty at McGill until the Faculty of Arts began teaching in 1843. This was followed by the establishment of the Faculty of Law in 1848, which is now the oldest law faculty in the nation. McGill's School of Architecture, founded in 1896, was the second of its kind in Canada, following the University of Toronto.

A significant figure in McGill's history is Sir John William Dawson, who served as principal from 1855 to 1893 and played a key role in transforming the school into a modern university. The campus itself is a testament to its rich heritage, featuring architectural landmarks like the West Wing of the Arts Building, designed by William Spier for William Molson in 1861, and major alterations to the East Wing by Alexander Francis Dunlop in 1888. George Allan Ross contributed several buildings, including the Pathology Building (1922–23) and the Neurological Institute (1933), with an addition in 1938. In 1934, Jean Julien Perrault designed the McTavish Street residence for Charles E. Gravel, now known as David Thompson House.
